Balchem’s Optifolin+ Demonstrates Strong Methylation Capacity: Study

The in vitro study found that the choline-enriched L-5-MTHF significantly upregulated methylation, which controls gene expression, influencing age-related biological outcomes.

Balchem’s Optifolin+, an L-5-methyltetrahydrofolate (L-5-MTHF) ingredient, was shown to significantly enhance cellular methylation capacity and DNA methylation in a human cell model study published in the Journal of Dietary Supplements. The findings provide mechanistic evidence of the ingredient’s biological activity, strengthening its scientific backing.

Healthy aging is increasingly being examined through the lens of epigenetics, which centers on how gene expression is regulated without altering the genetic code itself, Balchem reported. Gene expression is controlled by methylation, a cellular process in which carbon units (or methyl groups) are added to molecules such as DNA, influencing its function and expression. Disruptions in DNA methylation patterns are associated with genomic instability. Insufficient availability of methyl donor nutrients such as folate can contribute to genomic disruptions over time.

Optifolin+ was developed to support methylation through the synergistic combination of L-5-MTHF and choline, two methyl donors that work through interconnected one-carbon metabolic pathways. This unique pairing allows Optifolin+ to deliver as much as seven times greater methyl agents compared to other folate forms and achieve a 98% methyl capacity, optimizing support for DNA integrity and stability.

The new study, conducted in collaboration with the University of Arkansas for Medical Sciences, evaluated the biological activity of Optifolin+ in a human cell model (HepaRG) over a 96-hour period. The results showed that Optifolin+ modulated methyl-donor status, increasing S-adenosylmethionine (SAM) by 71% and enhancing cellular DNA methylation by up to 14 times.

“Folate plays a foundational role in cellular function, yet, modern lifestyle factors from stress or suboptimal diets can affect methylation efficiency. For brands looking to innovate beyond traditional folate products, understanding biological performance matters,” said  Lauren Eisen, senior marketing and business development manager for minerals and nutrients at Balchem’s human nutrition and health division. “This latest research demonstrates how Optifolin+’s unique choline-enriched structure translates into meaningful cellular activity. These findings provide a stronger scientific foundation for next-generation healthy aging and epigenetic health solutions.”
